  • Drug development to eradicate HIV/AIDS

Research Abstract:

I am developing drugs to eradicate HIV/AIDS based on high-resolution solid-state NMR structures determined with sensitivity enhanced EPR/NMR technology and methodology. My thesis research at MIT improved upon combined EPR/NMR technology that can increase the sensitivity of magic angle spinning (MAS) NMR experiments by a few orders of magnitude. MAS NMR yields atomic-resolution structural detail that is required to rationally design new analogs of existing drugs to increase binding affinity and specificity to elicit targeted biological activity. Our strategy is to combine experiments performed at cryogenic temperatures and lower magnetic fields (7 Tesla) with physiological temperatures experiments at high-field (20 Tesla) to yield structures of not only rigid bound ligands, but also information on dynamics and higher free-energy conformers. The eradication of HIV/AIDS is currently our highest priority, for which we are levering these new NMR structures to develop analogs of Protein Kinase C (PKC) activators that can activate latent HIV reservoirs.
